So in order to understand your power consumption, and thus your battery capacity, the first thing that you need to do is figure out how much current is drawn from the battery when traveling at 100 km/h. This will also be dependent upon the load, how flat the travel distance is (any hills?), and whether the surface is paved or not.
Knowing your average current draw from the battery will enable you to calculate the capacity of the battery for the range that you need.
Charging is a separate issue and is related to the capacity (C) of the battery (in Amp*hours, the maximum charge rate (which is usually the same as C), and how much life you want from the battery. Remember that fast charging will internally heat up the battery and will shorten the life of the battery as well as reduce its long term capacity.
